Protocol

BETA GALACTOSIDASE STAINING

PBS (Dulbecco's) 2% paraformaldehyde or | in PBS 0.5% glutaraldehyde 0.2 M K+ ferrocyanide 0.2 M K+ ferricyanide X gal - 40 mg/ml in DMSO (store frozen & use once) 1 M MgCl2
Remove media Wash lx with PBS if cells very adherent (optional) Add fixative - let sit 5 min. at room temperature Wash 3x with PBS Add staining solution. Need enough to cover cells. Final concentration: 1 mg/ml X gal 5 mM K+ ferrocyanide 5 mM K+ ferricyanide 2 mM MgCl2 PBS Let sit at 37°C 4 hr -- 16 hr. Read by light microscopy Can rinse cells with 3% DMSO in PBS and store at 4°C with PBS
